英文摘要Cellorganelleslikeperoxisomes,theendoplasmicreticulum(ER)andendosomeshavelongbeenconsideredasconfinedcompartmentswithdistinctisolatedfunctions.PeroxisomesparticipateinprimaryandsecondarymetabolismaswellasinReactiveOxygenSpecies(ROS)signaling,whereasendosomesserveasmultipurposeplatformsforlongdistancetransportofmacromolecules.However,recentadvancesshowthatorganellesareratherdynamicstructuresthatheavilyinteractwitheachother.Forexample,aclosemolecularlinkbetweenperoxisomesandendosomeswasestablishedinfungalcellsbyshowingthatmicrotubule-dependenttransportofearlyendosomesdistributesperoxisomesthroughouthyphae.Inaddition,lipiddroplets(LDs)andERsegmentsaretransportedbytheseendosomes.Atthesametime,theformationanddynamicsofLDs,endosomes,andperoxisomesdependontheER.Remarkably,mRNAsencodingseptinsareanadditionalimportantcargoforearlyendosomes.ExperimentaldatasupportamodelinwhichlocaltranslationofseptinmRNAsatthecytoplasmicsurfaceofendosomespromotesseptinoligomerassemblyandformationofhigher-orderstructures.Here,weproposetoinvestigate(i)thebiologicalfunctionofperoxisomesduringpolargrowthanddevelopment,(ii)apotentialfunctionallinkbetweenmRNAtransportandthecrosstalkofperoxisomes,ERandendosomes.ToprovideabroadviewontheseaspectsourteamconsistsoffungalbiologyexpertsusingestablishedmodelsystemssuchasUstilagomaydis,Aspergillusnidulans,NeurosporacrassaandPodosporaanserina.Genetic,biochemicalandcellbiologicalapproacheswillbecombinedtoidentifymolecularmechanismsandessentialcomponentsinorganelledynamics.Bycomparingthecrosstalkbetweenorganellesindifferentfungalmodels,wewilluncoverfundamental,evolutionaryconservedprocesses,aswellasspecificmechanismsadaptedtothelifestylesofdifferentfungi.Obtainedresultswillbeofgeneralinterestforcellbiologistsandthegainedknowledgecanbeappliedtoimprovetheproductionofsecondarymetabolitesandtodevelopnovelfungicides.
